چکیده

Investigations on nine different mammalian cell lines revealed that permanently growing cells of one morphological class have numerous membrane properties in common which are different or even lacking in the other cell class. With electrophysiological methods it is shown that the sensitivity of the Na<·K< pump to ouabain is three orders of magnitude higher in the ionically non· coupled epithelioid cells than in the ionically coupled fibroblastoid cells which respond like primary cultures. This is accompanied by considerably higher binding constants for ouabain of the epithelioid cells as was shown by [' HI ouabain binding and membrane potential measurements. The epithelioid cells also revealed a 50% lower relative amount of phosphatidylethanolamine and a GO-fold less net synthesis of phospatidylinositol. Finally, although primary cultures cannot proliferate without serum, permanent fibroblastoid cells have a reduced serum requirement and permanent epithelioid cells can proliferate without any serum.

Gap and low-resistance junctions between cells in culture.

In vitro cultured cells are connected by different membrane junctions which may serve as a matrix for the monolayer, but which may also enable intercellu lar communication. Some of these intercellular infor mation exchanges may be detected by ionic coupling between the cells through so-called “low-resistance junctions” . Gilula and cow orkers1, for instance, have shown that metabolic cooperat...

Mesenchymal Stem Cells Derived from Rat Epicardial Versus Epididymal Adipose Tissue

Objective(s) Some investigation has indicated that adipose-derived stem cells possess different surface epitopes and differentiation potential according to the localization of fat pad from which the cells were derived. In the present study proliferation capacity and aging of such cells were explored. Materials and Methods Adherent cells were isolated from the collagenase digests of adipose tiss...

ارزیابی سمیت سلولی سیس پلاتین، اکسید آرسنیک و استامینوفن روی خطوط سلولی سرطانی و نرمال

Introduction: Cell culture is a process in which the cells ware isolated from original tissue, dispersed in liquid media and then placed in culture plate where the cells adhere together and propagate. Today, this method is used for assessment of cell toxicity, its mechanisms and effect of different compounds on intracellular components. Methods: Clonogenic assay was used for assessment of cel...

O-13: Na+/K+-ATPase Alpha1 Isoform Mediates Ouabain-Induced Expression of Cyclin D1 and Proliferation of Rat Sertoli Cells

Background: Novel roles for the interaction of cardiotonic steroids to Na+/K+-ATPase have been established in recent years. The aim of the present study was to investigate the intracellular signaling events downstream the action of ouabain on Na+/K+-ATPase in Sertoli cell obtained from immature rats. Treatment of Sertoli cells with ouabain (1 μM) induced a rapid and transient increase in the ex...
